COOH-PLGA(20k)-PEG(5k)-MAL is a block copolymer which is reactive with thiol (SH) at pH 6.5-7.5 via formation of thioether linkage. The terminal carboxylic acid (COOH) can react with amine in the presence of activator, such as HATU or EDC, to form a stable amide linkage. The polymer can self-assemble in water with poly(lactide-co-glycolide) (PLGA) core and PEG shell arrangement. The core regions can serve as reservoir for hydrophobic drugs while the shell regions not only provide protection from degradation (enzymatic or hydrolysis) but also enables a stealth behavior for successful in vivo translation of nanomedicine.
