PROTAC linker is a crosslinker in the middle that connects two functional motifs of a PROTAC, a target protein binder and an E3 ligase recruiter. Most commonly used protac linkers in the development of PROTACs are PEGs, Alkyl-Chain and Alkyl/ether. Insertion of PEG increases the water-solubility of the molecules.
