5-Vinyl-uridine (VU) is a uridine derivative with a terminal alkene group used to monitor RNA transcription. VU is incorporated into new RNA by RNA polymerases, replacing endogenous uridine. VU-labeled RNA can be rapidly detected with high sensitivity via IEDDA cycloaddition between the vinyl group and labeled tetrazines. This allows detection through methods such as fluorescent microscopy or flow cytometry, providing insights into cellular transcription levels.
