ADC Payload

ADC Payload


ADC payload is a class of cytotoxic molecules that are conjugated to mAb (antibody) for oncology and other therapeutic treatment. The payload in the ADC conjugate will be released inside cells either through mechanisms of enzyme digestion, low pH, or reduction condition.

The payloads in clinically active ADCs are classified into four major classes:

  1. Tubulin Inhibitors - represented by Auristatin, Maytansine, and Tubulysin
  2. DNA damaging agents - represented by Calicheamicin, Duocarmycin, and IGN
  3. Topoisomerase (I & II) inhibitors - exemplified by SN-38, Exatecan, and DXd
  4. Immunomodulatory - exemplified by STING agonist, TLR8 agonist, and TLR9 agonist
